Determination of trastuzumab responsive microRNAs in SKBR3 and BT474 cells.

(Submitter supplied) To develop gene-miRNA and pathway-miRNA networks in trastuzumab treatment, we performed a recent miRNA microarray profiling in trastuzumab treated/untreated SKBR3 and BT474 cell lines. The cells were plated at a starting density of 2 millions in 100 mm cell culture dishes and then, they were treated with 6 ug/mL trastuzumab and PBS in two replicates. Total RNA was isolated with the TRIzol reagent (Invitrogen) according to the manufacturer's instructions.

Comparison of the functional microRNA expression in immune cell subsets of neonates and adults

(Submitter supplied) Diversity of biological molecules in newborn and adult immune cells contributes to differences in cell function and atopic properties. Micro RNAs (miRNAs) are reported involve in the regulation of immune system. Therefore, determining the miRNA expression profile of leukocyte sub-populations is important for understanding immune system regulation. In order to explore the unique microRNA profiling that contribute to altered immune in neonates, we comprehensively analyzed the functional miRNA signatures of eight leukocyte subsets (polymorphonuclear cells, monocytes, CD4+ T cells, CD8+ T cells, natural killer cells, B cells, plasmacytoid dendritic cells (pDCs), and myeloid dendritic cells (mDCs)) from both neonatal and adult umbilical cord and peripheral blood samples, respectively. more...

MicroRNA Expression Profiling of Saliva in Tongue Squamous Cell Carcinoma Patients

(Submitter supplied) 41% of all oral carcinomas have been found to localize at tongue, where they were characterized as being aggressive and having capacity to locally invade and relapse frequently. Despite considerable enhancements in the cancer diagnosis and treatment techniques, tongue squamous cell carcinoma (TSCC) still remains to be one of the most common and lethal cancer types in the head and neck region. In this study, we aimed to identify a signature of saliva-specific microRNAs (miRNAs) expression in TSCC patients. more...

The microRNAs with potential in predicting lung metastasis of triple negative breast cancer

(Submitter supplied) To identify lung metastasis associated microRNAs in triple negative breast cancer (TNBC), we have employed the commercially available Agilent Human miRNA V19.0 Microarray (Platform GPL19730) as a discovery platform. In comparison with LM-Normal, 11 microRNAs significantly altered in both LM-Met and LM-Tumor, and then three of them (hsa-miR-21-3p, hsa-miR-21-5p and hsa-miR-211-3p) were excluded, which were also up-regulated in RF-Tumor. more...

Engineering of a promoterless anti-viral RNAi hairpin into an endogenous miRNA locus [miRNA]

(Submitter supplied) Short hairpin RNA (shRNA) expression strategies that allow safe and persistent target mRNA knockdown are key to the success of many in vitro or in vivo RNAi applications. Here, we propose a novel solution which is expression of a promoterless miRNA-adapted shRNA (shmiRNA) from an engineered genomic miRNA locus. For proof-of-concept, we genetically “vaccinated” liver cells against a human pathogen, by using TALEns or CRISPR to integrate an anti-hepatitis C virus (HCV) shmiRNA into the liver-specific miR-122/hcr gene. more...

Salivary microRNAs May Be A Valuable Biomarker To Detect Resectable Pancreatic Cancer

(Submitter supplied) Early surgery is vital in the treatment of highly fatal pancreatic cancer (PC). But there is no valuable and non-invasive biomarker to screen PC currently. Studies showed many salivary molecules can detect several systemic diseases. We aimed to investigate whether salivary microRNAs (miRNAs) can act as a biomarker to detect resectable PC.
