Treatment protocol |
Malignant mesotheliomas were induced in Wistar rats by single peritoneal injection with MWCNTs and amosite asbestos suspended in artificial lung medium.

Tumors were collected from study animals. About half of the material was flash-frozen and stored in -80°C, while the remaining material was subjected to histopathological and immunohistochemical analyses.

Total RNA from tumors and control peritoneal tissues was isolated using the ‘AllPrep DNA/RNA Mini Kit’ (Qiagen), following manufacturer’s protocol.

The synthesis of double-stranded cDNA, the in vitro transcription, the cleanup of aRNA and biotin-labeling were undertaken with the 3´IVT Plus Kit (Thermo Fisher Scientific).

Hybridization protocol |
Biotinylated fragmented aRNA (10 µg ) was hybridized onto the GeneChip® Rat Genome 230 2.0 array. Hybridization was performed for 16 h at 60 rpm and 45°C in the GeneChip® Hybridization Oven 640. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

GeneChips were scanned using the GeneChip Scanner 3000 7G.
