Extraction protocol |
Visceral adipose tissued were stained by nile red and were collected by surgical manipulation under a fluorescence microscope. These tissue were stored in RNA-later (Ambion). Total RNA were extracted using RNeasy Mini Kit (Qiagen) and were quantified using a spectrophotometer (NanoDrop ND-100)

Three hundred nanograms of total RNA was converted into labeled cRNA using the Low RNA Input Fluorescent Linear Amplification Kit (Agilent) and Cy3-labeled cRNA (1.5 micrograms) were prepared according to manufacture’s protocol

Hybridization protocol |
1.5 ug of Cy3-labelled cRNA was fragmented following the manufacturers instructions. On completion of the fragmentation reaction, 250 ul of 2x Agilent hybridization buffer was added to the fragmentation mixture and hybridized to Agilent Zebrafish Whole Genome Oligo Microarrays (G4112A) for 17 hours at 65°C in a rotating Agilent hybridization oven. After hybridization, microarrays were washed 1 minute at room temperature with GE Wash Buffer 1 (Agilent) and 1 minute with 37°C GE Wash buffer 2 (Agilent), then dried immediately by air spray

The hybridized microarrays were scanned using a scanner (Agilent G2565BA)

The scanned images were analyzed with Feature Extraction software (Agilent). Bad probes were removed based on gIsFeatNonUnifOL,gIsPosAndSignif and gIsWellAboveBG
