Extraction protocol |
Total RNA was isolated from brain tissues (frozen or fresh) by a single-step method using TRIzol® Reagent (Invitrogen, USA). The RNA samples were treated with RNase-free DNase at 37oC for 20 min and stored at -80oC until further use. Total RNA (20 –50μg) isolated from tissues was cleaned up using RNeasy spin columns (Qiagen, Germany) before cDNA synthesis.

Biotinylated cRNA were prepared according to the Eukaryote Sample preparation as described in Expression Analysis Technical Manual, 2001, Affymetrix.

Hybridization protocol |
Following fragmentation, 20 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Rattus norvegicus Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.

Data processing |
The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 100.
