Plasma cells were obtained from the bone marrow aspirates by magnetic bead separation using the human CD138 RosetteSep kit (Stem Cell Technologies, Vancouver, British Columbia, Canada.The cells were stored in TRIZOL at −80°C until analysis. Total RNA was isolated from CD138+ sorted cells of myeloma patients using the miRVana RNA isolation kit according to manufactures’ instructions (Ambion, USA).

Briefly, 100 ng of total RNA was labeled with cyanine3-pCp using miRNA Complete Labeling and Hyb Kit (Agilent Technologies). Cy3-labeled samples were dried for up to 3 h in the vacuum concentrator and then resuspended in nuclease-free water.

Hybridization protocol |
Samples were hybridized onto Human miRNA 8X 60 K format microarrays (Agilent Technologies) based on miRBase Release 19.0 at 55 °C for 20 h.

Scan protocol |
After hybridization microarrays were washed with Gene Expression Wash Buffer Kit and scanned using SureScan microarray scanner D (Agilent Technologies).
