Treatment protocol |
Group A was treated with neural basal media + EGF + bFGF; Group B was treated with neural basal media + EGF +bFGF + IGF-1; Group C was cultured in neural basal media without any growth factor.

Growth protocol |
Both MSCs and MSCs-derived NPCs were cultured in humidified chamber at 37C with 5% CO2 supply.

Extraction protocol |
Total RNA extraction was performed using miRNeasy mini kit from Qiagen according to recommended protocol.

Biotinylated cRNA were prepared using FlashTag Biotin HSR RNA labeling kit according to the standard Affymetrix protocol from 100 ng total RNA (FlashTag Biotin HSR RNA Labeling Kit User Manual, 2013, Affymetrix).

Hybridization protocol |
Following biotinylation, 21.5 ul of cRNA were hybridized for 18 hr at 48C on miRNA GeneChip 2.0 Array in hybridization oven 645. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

Scan protocol |
GeneChips were scanned using GeneChip Scanner 3000 7G system.

Data processing |
Affymetrix Expression Console software v.1.2 was used for data summarization, normalization and quality control. Recommended processing algorithm RMA+DABG was applied.
