Treatment protocol |
Wheat heads at mid-anthesis were inoculated as follow: 10 µl of a 100,000 spores/ml suspension of the isolate Fusarium graminearum strain DAOM 180378 was point-inoculated into each of two basal florets of each developed spikelet of the wheat heads. Water was used as a mock-inoculation treatment. The inoculated plants were mist-irrigated for 10 second intervals every 15 minutes for 48 hours to maintain high humidity at a temperature of 25 oC. Five treated heads were used for each biological replicate, and inoculated spikelets were sampled at 1, 2 and 4 days after inoculation. The sampled tissues were immediately frozen in liquid nitrogen and stored at -80 C until used.

Plants were grown in growth cabinets at 20 C/16 hrs daylight; 16 C/8 hrs night until heads reach mid anthesis; then they were moved to the misting room.

Total RNA from each sample was isolated using a modified form of the guanidine isothiocyanate-cesium chloride method of Chirgwin et al (Biochem. 18: 5294-5299, 1979), as outlined in Ohan and Heikkila (Dev. Genet. 17: 176-184, 1995). The RNA samples were treated with an RNase-free DNase (Qiagen, Mississauga, Canada) and cleaned up using the RNeasy Mini Kit (Qiagen) according to manufacturer’s instructions.

Data processing |
The probe-level data for the whole profiling experiment were assembled into one dataset using the Robust Multichip Analysis (Irizarry et al, 2002) implemented in the software Acuity 4.0 (Molecular Devices, CA, USA), with quantile normalization and summarization by median polish.
