Treatment protocol Subsequently rats were randomly divided into Four groups (3 rats in each group): 1) non-stressed controls (C), 2) rats treated with Cortrosyn (ACTH) (10 IU) every 24h for 4 days, with the last injection on day 4 given 1h prior to harvesting of adrenal tissues, 3) rats treated with 17α-ethinyl estradiol (17α-E2), 10 mg/kg BW sc every 24h for 5 days, and 4) rats treated with dexamethasone (DEX), a single injection of 100 ug, sc for a 24 h period. At the end of treatment, animals were euthanized and adrenal tissues collected for various molecular and biochemical measurements.
Growth protocol Three monthes male Sprague Dawley (SD) rats were purchased from Harlan Laboratories (Indianapolis, IN). They were allowed to acclimatize to a new controlled environment (25 ± 2oC, 55 ± 5% relative humidity with a 12h light-dark cycle) for approximately one week.
Extracted molecule total RNA
Extraction protocol Total RNA was purified with miRNeasy Mini Kit (QIAGEN) according to the manufacturer's instructions.
Label biotin
Label protocol 500 ng total RNA containing low molecular weight RNA was labled with FlashTag™ Biotin HSR RNA Labeling Kit according to the standard Affymetrix protocol.
 
Hybridization protocol Affymetrix® GeneChip® Hybridization, Wash and Stain Kit was used for the hybridization according the standard protocal. GeneChips were washed and stained in the Affymetrix GeneChip Fluidics Station 450.
Scan protocol GeneChips were scanned using the Affymetrix GeneChip Scanner 3000 7G. The measurements were carried out by the Protein and Nucleic Acid (PAN) Microarray Facility of Stanford University.
Description miRNA expression data from rat adrenal gland treated with phosphate buffered saline (PBS).
Data processing The microarray data were analyzed using Partek® Genome Suite software, version 6.3 Copyright© 2008 (Partek Inc., St. Louis, MO, USA)
