Title |
Pig5_Failure |
Sample type |
RNA |
Source name |
Right ventricular free wall, heart failure
Organism |
Sus scrofa |
Characteristics |
pig number: Pig5 tissue: Right ventricular free wall breed: Mixed breed of Hampshire, Yorkshire and Swedish Landrace weight: 30-40 kg treatment group: Failure
Treatment protocol |
Heart failure induced by ligation of the coronary arteries on the free right ventricular wall. Heart failured was defined as right atrial pressure >20 mmHg.
Extracted molecule |
total RNA |
Extraction protocol |
Trizol extraction of total RNA was performed according to the manufacturer's instructions. RNA was further purified using RNeasy columns (Quiagen)
Label |
Biotin
Label protocol |
250 nanograms of total RNA from each sample were used to prepare biotinylated fragmented cRNA according to the GeneChip® 3’ IVT Express Kit Manual (PN 702646 Rev1, Affymetrix Inc., Santa Clara, CA)
Hybridization protocol |
Affymetrix GeneChip® expression arrays (Porcine Genome Array) were hybridized for 16 hours in a 45°C incubator, rotated at 60 rpm. According to the GeneChip® Expression Wash, Stain and Scan Manual (PN 702731 Rev2, Affymetrix Inc., Santa Clara, CA) the arrays were then washed and stained using the Fluidics Station 450
Scan protocol |
GeneChips were scanned using a GeneChip® Scanner 3000 7G
Description |
Gene expression data from acute ischemic heart failure sample
Data processing |
The raw data was processed using Partek Genomic Suite software (Partek Inc, St Louis, MO). Two-way analysis of variance (ANOVA) method was used to calculate p values.
