RNA library was prepared using tandem-affinity purification of a tagged protein covalently crosslinked to RNA. The 6xHis and Biotinylation site tags were used for protein purification. RNA was then prepared using a protocol adapted from Hafner et al (2010) for yeast (Jamonnak et al. submitted Mar 2011).
