Treatment protocol |
When high level engraftment was achieved the mice were randomised to receive either dexamethasone 15 mg/kg or vehicle control IP. Mice were culled at 0 hours, 8 hours, 24 hours or 48 hours following treatment. The mice in the 48 hour groups received a second dose of dexamethasone or vehicle control at 24 hours.

Growth protocol |
Cells from ALL-3, a glucocorticoid-sensitive xenograft, were inoculated by tail-vein injection into 28 NOD/SCID mice.

RNA was extracted using the RNeasy mini kit (Qiagen, Hilden, Germany) and the RNA integrity verified (Agilent Bioanalyzer, Santa Clara, CA).

The sample probe profiles with no normalisation or background correction were exported from BeadStudio (version 3.0.14, Illumina, San Diego, CA). The data were pre-processed using variance stabilising transformation and robust spline normalisation in lumi, from R/Bioconductor
