Treatment protocol |
Vc was added to mESC medium at a final concentration of 50 μg/ml. At D2 after the 2nd infection, transduced MEFs were changed to mESC medium with or without Vc. Vc was continuously added in the Vc treatment group.

Growth protocol |
MEFs were cultured in DMEM+10% FBS+NEAA+GlutaMax+penicillin/streptomycin (P/S); after two-rounds of infections, GFP- or SKO-transduced MEFs were cultured in mESC standard medium (DMEM+15% FBS+NEAA+GlutaMax+Sodium pyruvate+P/S+β-mercaptoethanol+1,000U/ml LIF) with or without Vc.

Biotinylated cDNA were prepared according to the standard Affymetrix protocol from 100 ng total RNA (Whole Transcript (WT) Sense Target Labeling Assay Manual, 2007, Affymetrix).

Hybridization protocol |
Following fragmentation and labeling, 10 ug of cDNA were hybridized for 16 hr at 45C on GeneChip MoGene 1.0 ST Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

Scan protocol |
GeneChips were scanned using the Affymetrix Scanner 3000 7G.
