Description |
Rice suspension cultured cells (Oryza sativa L. cv. Nipponnbare) used for total RNA extraction were maintained using a modified N-6 medium and incubated on a rotary shaker at 25 C and 120 rpm in a dark. Cells were transferred to a fresh medium every week. Those cells 4 or 5 days after transfer to the new medium were used experiment.40mg of cultured rice cells were transferred to 1 ml of the fresh medium in a 2 ml centrifuge tube and pre-incubated for 30 min at 25 C on a thermomixer shaken at 750 rpm. N-acetylchitooctaose elicitor (100nM) was added to the culture medium and incubated for 2hr.Total RNA was extracted from rice cells by RNeasy Plant Mini Kit (QIAGEN Inc., Valencia, CA, USA).
