Treatment protocol Cultured endothelial cells for 1 weeks produced cells at passage one (control cells). Serial passaging was performed on a weekly basis until cells at passage four (senescent cells) were reached (test1 cells). Regenerated endothelial cells (test2 cells) was obtained by regeneration after endothelial denudation in vivo, scraped and cultured without further passage. Both control and test cells at basal condition were collected for total RNA extraction for microarray analysis.
Growth protocol porcine coronary arterial endothelial cells were brought to primary culture. Cultured endothelial cells for 1 weeks produced cells at passage one (control cells). Serial passaging was performed on a weekly basis until cells at passage four (senescent cells) were reached (test cells). Regenerated endothelial cells (Reg cells) was obtained by regeneration after endothelial denudation in vivo, scraped and cultured without further passage.
Extracted molecule total RNA
Extraction protocol Total RNA was extracted with Rneasy mini kit (Qiagen) performed according to the manufacturer's instructions.
Label biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 0.5-1ug total RNA.
 
Hybridization protocol Following fragmentation, cRNA were hybridized for 16 hr at 45C on GeneChip Porcine Genome Array.
Scan protocol GeneChips were scanned using the Affymetrix GeneChip Scanner 3000.

Data processing The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method.
