Title |
Infection with Brucella abortus C24 strain for 12 h-1 |
Sample type |
RNA |
Source name |
RAW 264.7 cells
Organism |
Mus musculus |
Characteristics |
cell line: RAW 264.7 culture media: RPMI infection: Brucella abortus C24 strain time point: 12h
Treatment protocol |
Cells were infected with 10 MOI of Brucella abortus wild and mutant strains for 0, 6, 12, and 24 h.
Growth protocol |
8.0 x 104 cells/cm2 in 6-well plates with 2 mL of RPMI containing 2 % FBS
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was isolated from the cells using RNeasy Mini Kit (Qiagen, Valencia, CA, USA) according to the manufacturers’ protocol. After processing with DNase digestion, clean-up procedures, RNA samples were quantified, aliquot and stored at -80°C until use. Quality control was performed with Agilent Bioanalyser.
Label |
biotin
Label protocol |
The sense cDNA was then fragmented and biotin-labeled with TdT (terminal deoxynucleotidyl transferase) using the GeneChip WT Terminal labeling kit.
Hybridization protocol |
Approximately 5.5 μg of labeled DNA target was hybridized to the Affymetrix GeneChip Array at 45°C for 16hour
Scan protocol |
Hybridized arrays were washed and stained on a GeneChip Fluidics Station 450 and scanned on a GCS3000 Scanner (Affymetrix).
Description |
Leukemic monocyte macrophage cell line
Data processing |
Array data export processing and analysis was performed using Affymetrix® GeneChip Command Console® Software (AGCC)
