Treatment protocol At postnatal day 29, Long Evans Hooded rats were split into four groups (two treatment and two control groups). Two groups of rats were divided into pairs based on gender and body weight and weaned onto an ad lib liquid ethanol (6.7%)-containing diet or an isolcaloric liquid control diet (containing maltose instead of ethanol) for 3 weeks. The other two groups of rats began receiving the same liquid ethanol or control diet in young adulthood (postnatal day 79) using a 3 week episodic binge-drinking paradigm (3 days liquid ethanol or control diet followed by 4 days ad lib solid chow). After the three week ethanol exposure period, all rats in each group were euthanized with CO2 and blood was collected into BD Vacutainer® SST™ tubes for serum miRNA isolation.
