Treatment protocol The mice were divided randomly into three groups of eight to 12 mice each. The animals in the control group were intraperitoneally injected with 100 µL DMEM, whereas the two treated groups were intraperitoneally injected with 100 µL of DMEM solution containing insulin (40 µg/mL) or in situ biomineralization modified insulin (total insulin concentration was 40 µg/mL, in which 11% was in BI form).
Growth protocol The mice were fed with powdered chow consisting of 48% carbohydrates, 17% protein, 34% fat, 1% fiber, minerals, and vitamins (HFK Biosciencem, China), and water was given ad lib.
Extracted molecule total RNA
Extraction protocol Total RNA was extracted using TRIZol Reagent (Life technologies) and purified using RNeasy Mini kit (QIAGEN), following manufacturers’ protocols.
Label biotin
Label protocol Total RNA were amplified, labeled, and purified using GeneChip® 3’IVT Express Kit (Affymetrix) according to the manufacturer’s instructions to obtain biotin labeled cRNA.
 
Hybridization protocol Labeled cDNA was hybridized to the GeneChip Mouse Genome 430A 2.0 array (Affymetrix, Santa Clara, CA) at 45°C for 16 h. The arrays were washed and stained in the Affymetrix GeneChip Fluidics Station 450 using the supplier’s reagents.
Scan protocol Slides were scanned by GeneChip® Scanner 3000.
Description Type 2 diabetes
Data processing Slides were scanned by GeneChip® Scanner 3000 and Command Console Software 3.1 with default settings. Spike-in control transcripts were monitored to verify hybridization integrity.
