Treatment protocol The cells were collected from 10cm2 culture dishes using RLT lysis buffer from the Qiagen miRNeasy kit, were stored at -80°C until RNA extraction.
Growth protocol The cells were grown in DMEM High Glucose + stable Glutamine (EK 2 mM) (PAA #E15-883) + 1 % v/v AA (von PAA # P11-002 1% v/v) + 10 % FCS GOLD (PAA # A15-151)
Extracted molecule total RNA
Extraction protocol The RNA was extracted using the provider's recommendations for the Qiagen miRNeasy kit, the RIN was measured, only high quality RNA (RIN 9-10) was used for hybridisation.
Label Biotin
Label protocol FlashTag Biotin HSR RNA Labeling procedure as recommended by Affymetrix, as start material 1µg RNA was used.
 
Hybridization protocol Labelled cRNA was hybridized to Affymetrix GeneChip miRNA 2.0 Microarrays for 17h at 48 °C, stained by strepatavidin/phycoerythrin conjugate.
Scan protocol The chips were scanned as described in the manufacturers´ protocol (FlashTag™ Biotin HSR RNA Labeling Kit P/N 703095 Rev. 2).
Description miRNA expression in fibroblasts derived from female breast
Data processing The data were normalized using Robust Multiarray Average (RMA) within the R environment for statistical computing. Non-human miRNAs and miRNAs with intensity below 80 in all age groups were excluded. The GSE55299.txt contains only human miRNAs that passed the expression threshold.
