Sample type |
genomic |
|
|
Source name |
iMSC#3 , Day 28 osteogenic differentiaiton (June)
|
Organism |
Homo sapiens |
Characteristics |
cell line: iMSC#3 sample origin: Human bone marrow-derived mesenchymal stromal cell gender: Male treatment: osteogenic differentiaiton
|
Growth protocol |
The iMSC3 cells were culture in minimum essential medium alpha (MEM alpha) (Invitrogen) supplemented with 10% fetal calf serum (FCS) and glutamax (growth mediun; GM). For osteogenic differentiation GM was supplemented with 67 µM ascorbic acid 2-phosphate (Sigma-Aldrich), 3.5 mM β-glycerol phosphate (Sigma-Aldrich) and 10 nM dexamethasone (Sigma-Aldrich) (differentiation medium; DM) and cells were cultured for 28 days.

Extracted molecule |
genomic DNA |
Extraction protocol |
DNA was isolated using the Wizard Genomic DNA Purification Kit (Promega, Madison, USA).

Label |
Biotin and dinitrophenyl
|
Label protocol |
Standard IIlumina label protocol
|
|
|
Hybridization protocol |
Standard IIlumina hyb protocol
|
Scan protocol |
Standard IIlumina scan protocol
|
Data processing |
Data extraction and initial quality control of the bead summary raw data were performed using GenomeStudio v2011.1 and the Methylation module v1.9, both provided by Illumina.
