For prostate sphere differentiation: E-PZ-iPS-like cells cultured on Matrigel were digested with collagenase type IV (Invitrogen) and transferred to ultra-low attachment plates (Corning Life Sciences, Tewksbury, MA) for suspension culture for 8 days in DMEM/F12 (1:1) medium supplemented with 5 ng/ml bFGF (PeproTech), 10 ng/ml LIF (Santa Cruz Biotechnology Inc.), and 5% knockout FBS (Invitrogen). For basal epithelial cell differentiation, E-PZ-iPS-like spheres were then cultured in Complete PFMR-4A medium [29] for 3 days. To induce expression of androgen receptor (AR), E-PZ-iPS-like spheres were further cultured in Complete PFMR-4A medium with 10 nM R1881 for 3 days. To induce expression of prostate-specific antigen (PSA), E-PZ-iPS-like spheres were co-cultured with rat urogenital sinus (UGS) isolated from E17 rat embryos as previously described [31] in Complete PFMR-4A medium with 10 nM R1881 for 5 days.
