Thyroid carcinoma models

Analysis of 2, 6, and 10 month old transgenic (Tg) thyroids expressing RET/PTC3 fusion gene or E7 human papillomavirus 16 oncoprotein. The two Tg populations with different initiating mutations both develop thyroid carcinoma. Results provide insight into the different steps leading to carcinoma.

(Submitter supplied) The RET/PTC3 (RP3) fusion gene is the most frequent mutation found in radiation-induced papillary thyroid cancers (PTC). Several studies suggest that the RET/PTC rearrangement is an initiating event in tumorigenesis. E7 is an oncoprotein derived from the Human Papilllomavirus 16 (HPV16) responsible for most cervical carcinoma in women. The role of micro-RNA genes in papillary thyroid carcinoma

(Submitter supplied) We show that numerous miRNAs are transcriptionally up-regulated in papillary thyroid carcinoma (PTC) tumors compared with unaffected thyroid tissue. Among the predicted target genes of the three most upregulated miRNAs (miRs 221, 222 and 146b), only less than 15% showed significant downexpression in transcript level between tumor and unaffected tissue. The KIT gene which is known to be downregulated by miRNAs 221 and 222 displayed dramatic loss of transcript and protein in those tumors that had abundant mir-221, mir-222, and mir-146b transcript. more...

Papillary thyroid carcinoma

Analysis of papillary thyroid carcinoma (PTC) tumors from 9 patients. PTC accounts for about 80% of all thyroid cancers. These results, together with those obtained from custom miRNA chips, indicate the involvement of several miRNAs and the receptor tyrosine kinase KIT in PTC pathogenesis.

GENE EXPRESSION PROFILE REGULATED BY THE HPV16-E7 ONCOPROTEIN AND ESTRADIOL IN CERVICAL TISSUE

(Submitter supplied) The HPV16-E7 oncoprotein and 17β-estradiol are import factors for the induction of premalignant lesions and cervical cancer. The study of these factors is crucial for a better understanding of cervical tumorigenesis. In this study, we performed a microarray analysis to obtain a global gene expression profile induced by both, HPV16-E7 and 17β-estradiol in cervical tissue of K14E7 transgenic mice. We found that 17β-estradiol is the main cause of the up-regulation of a large number of cellular genes involved in the immune response whereas E7 oncoprotein mainly affects the cellular metabolism. more...
