NUDT7 is a member of the Nudix hydrolase family. Nudix hydrolases eliminate potentially toxic nucleotide metabolites from the cell and regulate the concentrations and availability of many different nucleotide substrates, cofactors, and signaling molecules (Gasmi and McLennan, 2001).
Gasmi and McLennan (2001) cloned 2 variants of mouse Nudt7, which they designated Nudt7-alpha and Nudt7-beta. Nudt7-alpha encodes a deduced 236-amino acid protein with a Nudix motif, a motif associated with CoA hydrolysis, and a C-terminal peroxisome targeting sequence. Nudt7-beta encodes a 216-amino acid protein that lacks a 20-amino acid stretch, including the C-terminal end of the Nudix motif, compared with Nudt7-alpha. RNA dot blot analysis of human tissues detected high to moderate expression in liver, lung, kidney, pancreas, pituitary, small intestine, spleen, and placenta. Expression in several brain regions was generally low. RT-PCR of mouse tissues detected ubiquitous expression of both Nudt7-alpha and Nudt7-beta. Fluorescence-tagged mouse Nudt7-alpha showed a punctate distribution in transfected HeLa cells, similar to the distribution of a peroxisome marker.
Gasmi and McLennan (2001) determined that mouse Nudt7-alpha is a CoA diphosphatase, and it hydrolyzed CoA, CoA esters, and oxidized CoA with similar efficiencies. It did not hydrolyze other common nudix hydrolase substrates. Activity was optimal at pH 8.0 in the presence of Mg(2+) or Mn(2+), and fluoride was inhibitory. Nudt7-beta was inactive against all substrates tested. Gasmi and McLennan (2001) concluded that the function of NUDT7 may be to eliminate oxidized CoA from peroxisomes or to regulate CoA and acyl-CoA levels in response to metabolic demand.
Hartz (2005) mapped the NUDT7 gene to chromosome 16q23.1 based on an alignment of the NUDT7 sequence (GenBank AA227330) with the genomic sequence.
