Genome-wide expression change by SHARPIN knockdown in MCF-7 cells

(Submitter supplied) We aim to investigate the role of SHARPIN in breast cancer progression. MCF-7 cells were used as the model and SHARPIN was silenced by siRNA.SHARPIN (SHANK-Associated RH Domain Interactor) is also named as SIPL1 and HSIPL1. The gene is located in Chr 8. SHARPIN protein is shown to regulate cell adhesion and migration. Besides, SHARPIN is proved to the component of the LUBAC complex and is necessary for NKFB function, while SHARPIN KO mice will lead to chronic dermatitis and immune deficiency in B cells.

(Submitter supplied) Ovariectomized virgin Ccnd1-/- and Ccnd1+/+ mice (5 weeks of age) were allowed to recuperate for 2 weeks. The mice were assigned to either replacement pellets containing E2 (0.75 mg, 60-day release) or pellet containing placebo. Mice were sacrificed at day 7 after pellet implantation. RNA extracted from mammary glands (3 each group) was labeled and used to probe Affymetrix 430_2.0 arrays.

Role of the COP1 E3 ligase in ERalpha signaling

(Submitter supplied) Our study defines the E3 ligase COP1 as a positive modulator of ERalpha-dependent gene transcription. We carried our RNA-Seq experiments with total RNAs from control or COP1-depleted MCF7 cells treated or not with estrogens and identified a signature of genes known to be regulated by estrogens whose transcription is deregulated upon COP1 deficiency.
