Normal mammary glands at three developmental stages: adipose stroma

Analysis of adipose stroma from nulliparous, lactating and involuting mammary glands. Pregnancy-associated breast cancers (PABCs) diagnosed during lactation are generally more aggressive with poorer prognosis. Results provide insight into the role of stromal changes in the pathogenesis of PABCs.

Expression Data from Mouse Mammary Gland Adipose Stroma

(Submitter supplied) Adipose stroma in the mouse mammary gland undergoes remodeling throughout the 5 stages of development. These include nulliparous (virgin;never been pregnant), pregnant, lactating, involuting and regressed. We used a microarry to determine the changes in gene expression during gland development.

mRNA Expression Comparison Between Igfbp7-null and wild type mice at Lactation Day 3

(Submitter supplied) Insulin like growth factor binding protein 7, Igfbp7, is a secreted protein that in addition to modulating insulin and insulin-like growth factor signaling, it acts as a tumor suppressor gene in breast and other cancers. To elucidate the role of Igfbp7 in regulating the proliferation and differentiation of mammary epithelial cells, we examined the growth and differentiation of mammary gland through different stages of its development in Igfbp7-null mice. more...

Stromal PTEN inhibits the expansion of mammary epithelial stem cells through Jagged-1

(Submitter supplied) Fibroblasts within the mammary tumor microenvironment are active participants in carcinogenesis mediating both tumor initiation and progression. Our group has previously demonstrated that genetic loss of PTEN in mammary fibroblasts induces an oncogenic secretome that remodels the extracellular milieu accelerating ErbB2-driven mammary tumor progression. While these prior studies highlighted a tumor suppressive role for stromal PTEN, how the adjacent normal epithelium transforms in response to PTEN loss was not previously addressed. more...

Pregnancy-activated fibroblasts of the mouse mammary gland

(Submitter supplied) The mouse mammary gland undergoes severe morphological changes during each pregnancy cycle. These are controlled by epithelial as well as stromal factors, including fibroblasts. This project aimed to identify factors that are expressed in mammary fibroblasts during early pregnancy (day3) when the first morphological changes become microscopically visible.

Gene expression profile of uniparous mouse mammary cell luminal progenitor subpopulation FACS-sorted based on expression of CD24CD29CD61

(Submitter supplied) We modeled two types of breastfeeding behaviors in wild type FVB/N mice: (1) normal or gradual involution of breast tissue following prolonged breastfeeding and (2) forced or abrupt involution following short-term breastfeeding. To accomplish this, pups were gradually weaned between 28 and 31 days (gradual involution) or abruptly at 7 days postpartum (abrupt involution). FACS was used to relatively quantify mammary epithelial subpopulations. more...

A cell transcriptomic profile reveals the molecular mechanisms of adipocytes in mammary gland across development

(Submitter supplied) Studying composition and developmental mechanisms in mammary gland is crucial for healthy growth of the newborn. Here, we constructed the largest transcriptomic dataset of mammary gland cells thus far. The dataset captured 126,829 high-quality nuclei from adipocytes, endothelial cells, epithelial cells, fibroblasts cells, immune cells, myoepithelial cells and precursor cells at five timepoint of mammary development. more...

Transcriptome of normal mammary glands of Sprague-Dawley rats at six stages of development: pre-pubertal, peri-pubertal, pubertal, lactation, adult parous and age-matched nulliparous

(Submitter supplied) There is a lack of systematic investigations of large-scale transcriptome patterns associated with normal breast development. Herein, we profiled whole-transcriptome (by microarrays) of normal mammary glands in female Sprague-Dawley rats, an animal model widely used in breast cancer research, across six distinctive developmental stages – pre-pubertal, peri-pubertal, pubertal, lactation, and adult parous and age-matched nulliparous.

Paternal obesity and epigenetic inheritance of breast cancer: The role of systemic effects and transmission to the second generation

(Submitter supplied) Mammary gland from F1 CO female offspring exhibited enhanced development when transplanted into OID females [OID(CO-MG)], as shown by higher mammary gland area, epithelial branching and epithelial elongation, compared to CO females that received a CO mammary gland [CO(CO-MG)]. Similarly, mammary tumors from F1 CO female offspring transplanted into OID females [OID(CO.T)] displayed improved growth with higher proliferation index and lower apoptotic rates. more...

Creatine mediated crosstalk between adipocytes and cancer cells regulates obesity-driven breast cancer

(Submitter supplied) Obesity is a major risk factor for adverse outcomes in breast cancer. Maguire, Ackerman et al. reveal Gatm and Acsbg1 as molecular regulators of obesity-driven breast cancer progression. They further show that in obesity creatine is a key metabolite in the crosstalk between adipocytes and breast tumors.

Effect of Cabergoline on Post-Lactational Involution and Long-Term Mammary Gland Behavior.

(Submitter supplied) The aim of this study is to assess the impact of cabergoline on the intensity of short-term post-lactational involution and to explore long-term alterations in the epithelial dynamics of the glandular component in Brca1/P53-deficient mice. This study also delves into the transcriptomic changes associated with these processes. To further investigate the molecular changes occurring at these temporal points, RNA sequencing (RNA-seq) was conducted 1 day after post-lactation involution and again at 60 days.
