Polycomb group protein CBX2 deficiency effect on XY and XX embryonic gonads

Analysis of E11.5 XY and XX embryonic gonads deficient for the Polycomb group protein CBX2. Animals lacking CBX2 exhibit defects in gonadal, adrenal and splenic development. Results provide insight into role of Cbx2 in gonadal sex differentiation.

Expression data from Mouse embryo

(Submitter supplied) Mice lacking the function of the PcG protein CBX2 (also known as M33) show defects in gonadal, adrenal, and splenic development. In particular, XY knockout mice develop ovaries but not testes, and the gonads are hypoplastic in both sexes. To clarify the molecular basis of the defects, RNA prepared from both wild type and KO gonads were subjected to microarray analyses.

CBX2 is required to stabilize the testis pathway by repressing Wnt signaling

(Submitter supplied) Purpose: Determine whether sex-determining genes are bivalent at the bipotential stage, poised between the testis and ovary fate, and whether H3K4me3 and H3K27me3 resolve into sex-specific patterns after sex determination, contributing to the canalization and stabilization of either the testis or ovary fate. Methods: XX and XY supporting cells of the gonad were FACS-purified before sex determination (at E10.5) and after sex determination (at E13.5), and submitted to ChIP-seq for H3K4me3, H3K27me3 and H3 as a means to normalize across cell populations. more...

Gonadal Identity in the Absence of pro-Testis Factor SOX9 and pro-Ovary Factor beta-catenin

(Submitter supplied) The main goal of our study is to identify the molecular events that determine the gonadal identity in mammals. Although testis and ovary arise from a common embryonic primordium, they represent outcomes of opposing fate determination. This decision to differentiate into a testis or an ovary hinges upon the balance between two antagonizing factors, pro-testis SOX9 and pro-ovary β-catenin. This microarray analysis led to the identification of the genes involved in the fate of XX and XY gonads in absence of SOX9 and beta-catenin

Sex reversal following the deletion of a single far upstream enhancer of Sox9.

(Submitter supplied) Purpose: In this study we employed unbiased, genome wide techniques to identify novel enhancers of Sox9 that may cause Disorders of Sex Development (DSDs) when disrupted in the mouse. Methods: We performed ATAC-seq on 60K FACS-purified gonadal cells before and after sex determination to map nucleosome depleted regions (NDRs) indicative of regulatory elements. We then selected 16 putative enhancers present in Sertoli cells. more...

Basic helix-loop-helix transcription factor Tcf21 is downstream target of male sex-determining gene SRY

(Submitter supplied) The cascade of molecular events involved in mammalian sex determination has been shown to involve the SRY gene, but specific downstream events have eluded researchers for decades. The current study identifies one of the first direct downstream targets of the male sex-determining factor SRY as the basic-helix-loop-helix (bHLH) transcription factor TCF21. SRY was found to directly associate with the Tcf21 promoter SRY/SOX9 response element both in vitro and in vivo during male sex determination. more...

Cell type-specific role of CBX2-cPRC1 at the onset of spermatogonial differentiation [RNA-seq]

(Submitter supplied) Polycomb group (PcG) proteins maintain the repressed state of lineage-inappropriate genes and are therefore essential for embryonic development and adult tissue homeostasis. One critical function of PcG complexes is modulating chromatin structure. Canonical Polycomb repressive complex 1 (cPRC1), particularly its component CBX2, can compact chromatin and phase separate in vitro. These activities are hypothesized to be critical for forming a repressed physical environment in cells. more...

Identification of the transcription start site of transcribed sequences from the mouse Sry locus by CAGE-seq

(Submitter supplied) The mammalian sex-determining gene Sry induces male development. Through analyzing transcriptome of pre-Sertoli cells, we identified a novel sequence transcribed within palindromic sequence surrounding Sry. To examine whether this sequence contains transcription start site, Cap analysis gene expression (CAGE) -seq analysis was performed, in which putative transcriptional start sites can be identified by sequencing the 3’ end of cDNA corresponding to 5’ end of RNA. more...

Identification of a novel transcriptional variant in the mouse Sry locus by long-read RNA-seq and conventional RNA-seq

(Submitter supplied) The mammalian sex-determining gene Sry induces male development. Sry is exclusively expressed in Nr5a1-highly expressing gonadal supporting cell precursor, pre-Sertoli cells, with a peak at embryonic day 11.5 (E11.5). We developed a method to enrich pre-Sertoli cells from mouse embryonic gonads carrying Nr5a1-hCD271 (LNGFR) transgene. To examine mRNA expression in pre-Sertoli cells, pre-Sertoli cells were purified and introduced into long-read RNA-seq and conventional RNA seq analyses. more...
