ATP effect on acute myeloid leukemia lymphoblasts

Analysis of lymphoblasts from 12 acute myeloid leukemia (AML) patients cultured for 24hrs with ATP. Extracellular nucleotide ATP inhibits proliferation and accumulation of AML cells. Results provide insight into the molecular mechanisms underlying the inhibitory activity of ATP on leukemic cells.

Expression data from untreated and ATP treated AML blasts

(Submitter supplied) In the present study, we investigated whether, and to what extent, P2Rs and their ligands are involved in the regulation of AML cells. Our findings show that AML blasts express several receptors belonging to the P2X and P2Y family. Although different samples respond differently to ATP and UTP stimulation (reflecting the variability intrinsic to the group of acute myeloid leukemias), all the tested samples appear to be responsive to purinergic signalling, as demonstrated by intracellular calcium mobilization. more...

CD34+ and Treatment with UTP and CXCL12

(Submitter supplied) Homing and engraftment of hematopoietic stem cells (HSCs) to the bone marrow (BM) involve a complex interplay between chemokines, cytokines, and non-peptide molecules. Extracellular nucleotides and their cognate P2 receptors are emerging as key-factors of inflammation and related chemotactic responses. In this study, we investigated the activity of extracellular adenosine-triphosphate (ATP) and uridine-triphosphate (UTP) on CXCL12-stimulated CD34+ HSC chemotaxis. more...

UTP and alpha-chemokine CXCL12 effect on CD34+ hematopoietic stem cells

Analysis of CD34+ hematopoietic stem cells (HSCs) treated with UTP, alpha-chemokine CXCL12, or both. CXCL12 is a HSC chemoattractant secreted by the bone marrow. Results provide insight into the molecular mechanisms underlying the enhancement of CXCL12-dependent HSC migration by UTP.

Gene expression profiling of CD34+ subsets in Multiple Myeloma and healthy individuals

(Submitter supplied) Multiple myeloma (MM) is a clonal plasma cell disorder frequently accompanied by hematopoietic impairment. Genomic profiling of distinct HSPC subsets revealed a consistent deregulation of signaling cascades, including TGF beta signaling, p38MAPK signaling and pathways involved in cytoskeletal organization, migration, adhesion and cell cycle regulation in MM patients.

hMSC ATP treatment

(Submitter supplied) Nucleotides triphosphates are extracellular messengers binding to specific plasma membrane receptors (P2Rs) that modulate responses as different as proliferation, differentiation, migration or cell death on several cell types including hematopoietic stem cells. Little and controversial information is available on the role of extracellular nucleotides in human mesenchimal stem cells (hMSCs). In this study, we assessed whether P2Rs are expressed and functional in bone marrow-derived hMSCs. more...

SDF-1 induced transcriptional changes in human cord blood CD34+ cells

Thrombopoietin (TPO) protects against heart damage induced by doxorubicin and infarction in rat models

(Submitter supplied) In order to examine the mechanism of TPO on cardiac protection against DOX damage, we have employed whole genome microarray expression profiling as a discovery platform to identify genes with the potential to delineate the TPO cardioprotective mechanism against DOX-induced cardiomyopathy

Gene expression profiles of colorectal cancer cells grown in 2D/3D conditions

(Submitter supplied) Gene expression analysis was performed through RNA-Sequencing on samples prepared from a panel of four CRC cell lines (HT29, HCT116, LS174T, LS513) grown in either 2D or 3D cell culture conditions. Our gene expression data reveal that more than 2000 gene expression profiles are affected by the growth conditions for each cell lines even if samples prepared from each cell line still group together. Our data also demonstrate that hierarchical clustering mostly relies on the MSS/MSI phenotypes of the CRC cells.

Gene expression profiling of LT-HSCs and ST-HSCs conditionally expressing MLL-ENL

(Submitter supplied) We have developed a new conditional transgenic mouse showing that MLL-ENL, at an endogenous-like expression level, induces leukemic transformation selectively in LT-HSCs. To investigate the molecular mechanism of leukemic transformation in LT-HSCs conditionally expressing MLL-ENL, we preliminarily performed comprehensive gene expression profiling of CreER-transduced LT-HSCs and ST-HSCs using cDNA microarray analysis.

Expression data from human hematopoietic stem and progenitor compartments from patients with acute myeloid leukemia with monosomy 7 and healthy controls

(Submitter supplied) We applied a novel approach of parallel transcriptional analysis of multiple, highly fractionated stem and progenitor populations in a genetically defined subset of AML (AML with monosomy 7). We isolated phenotypic long-term HSC (LT-HSC), short-term HSC (ST-HSC), and committed granulocyte-monocyte progenitors (GMP) from individual patients with AML, and measured gene expression profiles of each population, and in comparison to their phenotypic counterparts from age-matched healthy controls.

Chronic myelogenous leukemia hematopoietic stem cells

(Submitter supplied) We show the molecular and functional characterization of a novel population of lineage-negative CD34-negative (Lin- CD34-) hematopoietic stem cells (HSCs) from chronic myelogenous leukemia (CML) patients at diagnosis. Molecular caryotyping and quantitative analysis of BCR/ABL transcript demonstrated that about one third of CD34- was leukemic. CML CD34- cells showed kinetic quiescence and limited clonogenic capacity. more...
