Spermatogenesis

(Submitter supplied) A multitude of genes expressed solely in meiotic or postmeiotic spermatogenic cells offers a myriad of contraceptive targets. Understanding mammalian spermatozoan development and the events surrounding fertilization has grown slowly, in part because of uncertainty about the number and identity of the cellular components involved. Determination of those transcripts expressed specifically by germ cells should provide an inclusive list of probable critical proteins. more...

High-Resolution Profiling of Novel Transcribed Regions During Rat Spermatogenesis

(Submitter supplied) Mammalian spermatogenesis is a complex and highly orchestrated combination of processes in which male germline proliferation and differentiation result in the production of mature spermatozoa. If recent genome-wide studies have contributed to the in-depth analysis of the male germline protein-coding transcriptome, little effort has yet been devoted to the systematic identification of novel unannotated transcribed regions expressed during mammalian spermatogenesis. more...

Expression profiling of rainbow trout testicular development and spermatogenesis

(Submitter supplied) Trout spermatogenesis proceeds seasonally in a way that all morphological and cellular events tend to be synchronized. We thus used gonads at key stages of the male reproductive cycle together with isolated germ cell populations to study the changes in gene expression underlying testis development. Statistical analysis followed by clustering of expression profiles allowed the discrimination of sequential events of gene activation or repression during testis development and/or throughout the germline.

Small RNA and mRNA transcriptome profiling in wildtype mice at multiple stages of spermatogenesis, from whole testis at day 1 postpartum (pp), day 3 pp, and day 7 pp and purified leptotene/zygotene and pachytene spermatocytes.

(Submitter supplied) MicroRNAs are essential for spermatogenesis. However, the stage‑specific requirements for specific miRNAs in the male mammalian germ line remain largely uncharacterized. The miR‑34 family is, to date, the only miRNA proven to be necessary for the production of sperm in mice, though its germline roles are poorly understood. Here, we generate and analyze paired small RNA and mRNA profiles across different stages of germline development in male mice, focusing on time points shortly before and during meiotic prophase I.

Developmental changes in RNP and Polysome associated mRNAs in Mouse testes

(Submitter supplied) Gametes rely heavily on post-transcriptional control mechanisms to regulate their differentiation. In eggs, the storage and selective temporal activation of maternal mRNAs is essential for normal development. In the male, transcription ceases during spermiogenesis necessitating the post-transcriptional regulation of many paternal mRNAs required for spermatid differentiation and spermatozoan function. more...

Ribonucleoprotein- and polysome-associated mRNAs of the testis: time-course

Analysis of ribonucleoprotein (RNP) and polysome fractions of prepuberal and adult testes. Protein synthesis occurs on polysomes and translationally inactive mRNAs are sequestered as RNPs. Results provide insight into the translation state of individual mRNAs as spermatogenesis proceeds.

Microarray profiling of total testicular transcriptome in mouse

(Submitter supplied) Gene expression during spermatogenesis is highly variable and this differential pattern is very important for the successive culmination of different stages of the process, leading to production of the male gamete. Taking the time window of first wave of spermatogenesis, we did a microarray profiling of total testicular transcriptome in mouse and found several significant patterns of variable gene expression, forming upregulated and downregulated clusters among the three stages analyzed here.

Microarray profiling of total testicular microRNAs in mouse

(Submitter supplied) Gene expression in mammalian testis undergoing spermatogenesis is under strict post transcriptional regulation and microRNAs are known to play a role in this regulation. Considering the time window of first wave of spermatogenesis, we did a microarray profiling of total testicular microRNAs in mouse and found several significant patterns of variable expression of these molecules during the different stages analyzed here.

Drosophila melanogaster spermatogenesis expression profile: mitotic, meiotic and post-meiotic cells

(Submitter supplied) We conducted a genome-wide expression analysis of wild-type males using three cell populations isolated from mitotic, meiotic and post-meiotic phases of spermatogenesis in Drosophila melanogaster. Our approach was to directly isolate testis regions enriched with RNAs from each of the three specific germline phases. We used microarrays to detail the global gene expression profile in spermatogenesis and identified up- and down-regulated genes between two different spermatogenic phases in pairwise comparisons

Transcriptomic sequencing of the murine testis during the first wave of spermatogenesis

(Submitter supplied) Our experimental design includes samples at different time points during the first wave of spermatogenesis. Each time point corresponds to specific cell content in the testis. First time point is post natal day (PND) 7, when only somatic cells and spermatogonia are present in the testis. Thereafter additional cell types are present in selected samples in chronological order: PND 14 contains early spermatocytes, PND 17 late spermatocytes, PND 21 round spermatids and finally PND 28 elongating spermatids. more...

Translational activation of developmental mRNAs during neonatal mouse testis development

(Submitter supplied) The sequence of gene regulatory events that drive neonatal germ cell development in the mammalian testis is not yet clear. We assessed changes in mRNA utilization in the neonatal testis at 1 and 4 dpp, times when the testis contains quiescent gonocytes (1 dpp) and proliferating spermatogonia (4 dpp). There are not thought to be major changes in the nature or number of somatic cells over that interval. more...
