T-dependent and T-independent germinal center B cells

Comparison of T-dependent (TD) and T-independent (TI) germinal center B cells from animals 4 days after immunization. The TD animals were carrier-primed 5 weeks earlier so that T-cell help was not limiting. Results provide insight into molecular differences between TD and TI germinal center B cells.

Comparative gene expression profiles of T-dependent and T-independent germinal centre B cells in mice

(Submitter supplied) Selection of B cells subjected to hypermutation in germinal centres (GC) during T-dependent (TD) antibody responses yields memory cells and long-lived plasma cells that produce high affinity antibodies biased to foreign antigens rather than self-antigens. GC also form in T-independent (TI) responses to polysaccharide antigens but failed selection results in GC involution and memory cells are not generated. more...

Germinal-center development of memory B cells driven by IL-9 from follicular helper T cells

(Submitter supplied) A subset of GC B cells that have stopped cycling, upregulated CD38 and downregulated BCL-6 is functionally verified as GC-derived memory B cell precursors (GC-MPs). RNA-seq analyses of the transcriptome were used to probe the developmental trajectory of these cells and their responses to IL-9, a cytokine that is found to drive the memory development from the GC.

Plexin B2 and Semaphorin 4C guide T-cell recruitment and function in the germinal center

(Submitter supplied) Follicular T-helper (TFH) cells are essential for germinal center (GC) responses. TFH localization in GCs is controlled by chemo-guidance cues and antigen-specific adhesion. Here we define an antigen-independent, contact-dependent, adhesive guidance system for TFH cells. Unusual for amoeboid cell migration, the system is composed of transmembrane plexin B2 (PlxnB2) molecule that is highly expressed by GC B cells and its transmembrane binding partner semaphorin 4C (Sema4C) that is upregulated on TFH cells. more...

Germinal center T follicular helper cell IL-4 production is dependent on SLAM receptor (CD150)

(Submitter supplied) CD4 T cell help is critical for both the generation and maintenance of germinal centers, and T follicular helper (TFH) cells are the CD4 T cell subset required for this process. SAP (SH2D1A) expression in CD4 T cells is essential for germinal center development. However, SAP-deficient mice have only a moderate defect in TFH differentiation as defined by common TFH surface markers. CXCR5+ TFH cells are found within the germinal center as well as along the boundary regions of T/B cell zones. more...

Expression data from healthy human PB B cell subsets

(Submitter supplied) Human PB B cell subsets are functionally distinct and may derive from different developmental pathways, reflected by their differential gene expression profiles. We used microarrays to detail the global programme of gene expression underlying germinal center transition and functional propensities of individual subsets according to surface receptor and cell adhesion molecule expression.

Conditional deletion of Bcl6 in naive B cells

(Submitter supplied) Bcl6 germline deletion causes a prominent inflammatory disease, owing to over-expression of Th2 cytokines, and affects the properties of B cells prior to immunization. Therefore we established the B cell-specific Bcl6 deletion mice and analyze the gene expression of naive B cells under physiological conditions.

Gene expressions of memory B cells and other type of B cells

(Submitter supplied) To obtain insight into the genetic basis of the increase of functional activity of memory B cells over time, we compared the gene expression profiles of day 7 and day 40 NP-specific/IgG1 memory B cells, GC B cells and plasma cells in immunized WT mice and naïve B cells, before and after activation in vitro.

The transcription factor Hhex cooperates with the corepressor Tle3 to promote memory B cell development

(Submitter supplied) Memory B cells (MBCs) are essential for long-lived humoral immunity. However, the transcription factors (TFs) involved in MBC differentiation are poorly defined. Here, by single cell RNAseq analysis, we identified a population of germinal center (GC) B cells in the process of differentiating into MBCs. Using an inducible Crispr/Cas9 screening approach we identified the hematopoietically-expressed homeobox gene Hhex as a transcription factor regulating MBC differentiation. more...

Single-cell genomics predict germinal centre-associated etiology of autoimmune risk loci

(Submitter supplied) The germinal centre (GC) response is critical for both effective adaptive immune responses and establishing peripheral tolerance by limiting auto-reactive B cells. To understand the gene regulatory principles underlying B cell maturation and the GC response we generated a single-cell transcriptomic and epigenomic atlas of the human tonsil, a widely studied, representative lymphoid tissue. We identify diverse immune cell subsets to help us map dynamic transcription factor activity during B cell activation, GC formation and plasma cell differentiation. more...
