Aging lungs and genetic background

Analysis of lungs of C57BL/6J and DBA/2J animals at up to 26 months of age. Aging lungs of C57BL/6J and DBA/2J strains differ in physiology and morphometry. Results provide insight into the influence of genetic background on the variability of age-dependent changes in lung structure and function.

Comparative analysis of gene expression in C57BL/6J and DBA/2J aging lungs.

(Submitter supplied) We hypothesize that gene expression in the aging lungs of these two strains of mice are divergent thus contributing to the disparity in the phenotypes. More specifically, (1) Aging DBA/2J mice compared to aging C57BL/6 mice are known to be accelerated in their lung physiology and morphometry; (2) C57BL/6J are known to have longer natural longevity than DBA/2J mice. In order to test these hypotheses at the gene expression level, we utilized microarray analysis to examine transcriptional differences between aging lungs of both strains of mice. more...

RNA sequencing profiling of the retina in C57BL/6J and DBA/2J mice: enhancing the retinal microarray datasets from GeneNetwork

(Submitter supplied) Purpose: The goal of the present study is to provide an independent assessment of the retinal transcriptome signatures of the C57BL/6J (B6) and DBA/2J (D2) mice and to enhance existing microarray datasets for accurately defining the allelic differences in the BXD recombinant inbred strains. Methods: Retinas from both B6 and D2 mice (3 of each) were used for the RNA-seq analysis. Transcriptome features were examined for both strains. more...

Evaluating Gene Expression in C57BL/6J and DBA/2J Mouse Striatum Using RNA-Seq and Microarray

(Submitter supplied) C57BL/6J (B6) and DBA/2J (D2) are two of the most commonly used inbred mouse strains in neuroscience research. However, the only currently available mouse genome is based entirely on the B6 strain sequence. Subsequently, oligonucleotide microarray probes are based solely on this B6 reference sequence, making their application for gene expression profiling comparisons across mouse strains dubious due to their allelic sequence differences, including single nucleotide polymorphisms (SNPs). more...

Microarray Analysis of Retinal Gene Expression in the DBA/2J Model of Glaucoma

(Submitter supplied) Purpose: The DBA/2J mouse is a model for secondary angle-closure glaucoma due to iris atrophy and pigment dispersion, which ultimately leads to increased intraocular pressure (IOP). We sought to correlate changes in retinal gene expression with glaucoma-like pathology by performing microarray analysis of retinal RNA from DBA/2J mice at 3 months before disease onset, and at 8 months, after IOP elevation. more...

DBA/2J model of glaucoma: retina

Analysis of retinas from strain DBA/2J with elevated intraocular pressure (IOP). A model for glaucoma, DBA/2J has mutations that lead to iris atrophy and pigment dispersion. These conditions result in increased IOP. Results provide insight into the molecular changes associated with IOP elevation.

Cystic fibrosis transmembrane conductance regulator deficiency in different genetic backgrounds

Analysis of lungs of C57BL/6J (B6) or BALB/c (BALB) animals deficient for cystic fibrosis transmembrane conductance regulator (CFTR). B6 background animals have a higher propensity to develop CF lung disease. Results suggest the variable severity of CF is controlled by multiple genetic factors.

Global transcriptome analysis reveals aging-related genes in Atp8b1 mutant lungs

(Submitter supplied) Aging is associated with progressive decline in lung function and responses to environmental stress culminating in increased incidence of lung diseases. Initial analysis of Atp8b1 (ATPase, Aminophospholipid Transporter, Class I, Type 8B, Member 1) mutant mice (on C57BL/6J background) revealed deficiency in transporter function and was associated with decreased lung function. Therefore, to further study the aging-related genes in the lungs at the molecular level, the transcriptome analysis of Atp8b1 mutant and C57BL/6J lungs were carried out at 14 M and 7_9 wks of age. more...

Gene expression profile of the age-related hearing loss cochlea in DBA/2J mice

(Submitter supplied) Age-related hearing loss (AHL) is the progressive loss of auditory function with aging. The DBA/2J (DBA) mice have been used as a model of AHL and undergoes progressive, age-related hearing loss by 12 weeks of age. Here we analyzed cochlear gene expression of 7-week-old and 36-week-old DBA mice using microarrays. Auditory brainstem response (ABR) analysis confrimed that severe age-related hearing loss occured in 36-week-old mice, whereas moderate hearing loss occured in 7-week-old mice. more...

Transcriptomic Analysis of the Hippocampus from Six Inbred Strains of Mice Suggests Basis for Sex-Specific Susceptibility and Severity of Neurological Disorders

(Submitter supplied) Identifying sex differences in gene expression within the brain is critical for determining why multiple neurological and behavioral disorders differentially affect males and females. Several are more common or severe in males (e.g., autism and schizophrenia) or females (e.g., Alzheimer’s disease and depression). We analyzed transcriptomic data from the mouse hippocampus of six inbred strains (129S1/SvImJ, A/J, C57BL/6J, DBA/1J, DBA/2J and PWD/Ph), to provide a perspective on differences between male and female gene expression. more...

Age-Dependent Differences in Gene Expression in Response to Calvarial Injury

(Submitter supplied) Children less than 2 years of age are capable of healing large calvarial defects, whereas adults have been found to lack this endogenous ability. In this study, we used microarray analysis to compare genomewide expression patterns during active regeneration after injury with calvaria in skeletally immature and mature mice. Parietal bone defects were created in 6-day-old (juvenile) and 60-day-old (adult) mice using a 4-mm trephine bit (n = 20 mice per age group). more...

Comparative analysis of gene expression in A/J CB versus DBA/2J

(Submitter supplied) We hypothesize that gene expression in the CBs of these two strains of mice are divergent thus contributing to the disparity in the phenotypes. More specifically, (1) genes related to CB function are expressed less in the A/J mice compared to DBA/2J mice; (2) gene expression levels of morphogenic and trophic factors of the CB maybe significantly lower in the A/J mice than the DBA/2J mice. In order to test these hypotheses, we utilized microarray analysis to examine transcriptional differences between CBs of both strains of mice. more...

Exon and junction microarrays detect widespread mouse strain- and sex-bias expression differences

(Submitter supplied) 71 liver RNA samples from three mouse strains - DBA/2J, C57BL/6J and C3H/HeJ - were profiled using a custom-designed microarray monitoring exon and exon-junction expression of 1,020 genes representing 9,406 exons. Gene expression was calculated via two different methods, using the 3'-most exon probe ("3' gene expression profiling") and using all probes associated with the gene ("whole-transcript gene expression profiling"), while exon expression was determined using exon probes and flanking junction probes that spanned across the neighboring exons("exon expression profiling"). more...

Genome wide DNA methylation profiling of whole blood samples from C57BL/6 and DBA/2J healthy mice

(Submitter supplied) Aging of mice can be tracked by DNA methylation changes at specific sites in the genome. In this study, we used the Infinium Mouse Methylation BeadChip to compare such epigenetic modifications in C57BL/6 (B6) and DBA/2J (DBA) mice. There were marked differences in age-associated DNA methylation in these commonly used mouse strains. In B6 the age-associated DNA methylation revealed general age-associated hypomethylation with focused hypermethylation at CpG islands, whereas this was hardly observed in DBA mice. more...
