The role of DNMT3A in AITL

(Submitter supplied) To investigate the role of DNMT3A in AITL, single cell RNA-seq (scRNA-seq) is performed to analysis the lymph nodes generated from the control, RHOAG17V and RHOAG17V DNMT3AR882H AITL mice. The mitoQC maps shows the distributions of unique gene detected and mitochondrial genes detected in each single cell. After the quality control, a total of 23,805 cells (7576 of control, 7913 of RHOAG17V and 8316 of RHOAG17V DNMT3AR882H) are used for clustering and further analyzing

Transcriptomic profiles of CD4+ T lymphocytes from RhoA wild type (WT) and RhoA G17V mice were generated by deep sequencing in duplicate, using Illumina poly-A RNA seq.

(Submitter supplied) The goal of this study is to identify transcriptomic effects of RhoA G17V expression in mouse CD4+ T lymphocytes. To this end, mice conditionally expressing RhoA G17V were generated by replacing exon 3 of RhoA with a minigene containing RhoA exons 3 to 6 flanked by loxP sites followed by a neomycin selection cassette and a mutant Rhoa exon 3 encoding the G17V substitution. Mouse model of RHOA G17V mutation in Peripheral T-Cell Lymphoma

Angioimmunoblastic T-cell lymphoma-like lymphadenopathy in mice transgenic for human RHOA with p.Gly17Val mutation

(Submitter supplied) A missense mutation in RHOA encoding p.Gly17Val has been reported to occur frequently in angioimmunoblastic T-cell lymphoma (AITL). Here, we describe a murine model which expresses the human RHOA mutant gene product in a T-cell specific manner and develops AITL-like symptoms. Gene expression profiles of lymphoma cells in homozygous Tet2 gene trap mice and CD4+ control cells.

(Submitter supplied) TET2 is an enzyme for converting methylcytosine (mC) to hydorxymethylcytosine (hmC) and its mutations have been frequently found in myeloid malignancies and T-cell lymphoma in humans. We analyzed Tet2 gene trap mice and found that homozygous mice developed T-cell lymphoma with follicular helper T-cell-like features. To examine comprehensive gene expression patterns of lymphoma cells, We performed gene expression analysis using lymphoma cells in homozygousTet2 gene trap mice and CD4+ control cells.
