Transcriptome profiling of normal breast tissue

(Submitter supplied) The Susan G. Komen Tissue Bank at Indiana University Simon Comprehensive Cancer Center (KTB) is the only repository of normal breast tissues donated by healthy women. Here, we examined the transcriptome profiling of normal breasts from 190 healthy women, including 62 African Americans, 11 Asians, and 109 Caucasians. Correlation analysis between geen expression and risk factors for breast cancer (such as smoking, BMI, age, racial background) was performed.

Analysis of microbiota composition within breast tumor and healthy breast tissues in males and females

(Submitter supplied) Total DNA was extracted from FFPE specimens of breast tumor and surrounding healthy tissue, amplified to collect amplicons of variable V3–V4 regions of the bacterial 16s rRNA gene and sequenced with MiSeq (2x300bp) Illumina platform.

Distinct cervical tissue-adherent and luminal microbiome communities correlate with mucosal host gene expression and protein levels in Kenyan sex workers

(Submitter supplied) Background: While the luminal microbiome composition in the human cervicovaginal tract has been defined, the presence and impact of tissue-adherent ectocervical microbiota remain incompletely understood. Studies of luminal and tissue-associated bacteria in the gastrointestinal tract suggest that they may have distinct roles in health and disease. Here, we performed a multi-omics characterization of paired luminal and tissue samples collected from a clinically well-characterized cohort of Kenyan women. more...

Multi-omics analysis of the cervical epithelial integrity of women using depot medroxyprogesterone acetate

(Submitter supplied) Depot medroxyprogesterone acetate (DMPA) is an injectable hormonal contraceptive used by millions of women worldwide. However, experimental studies have associated DMPA use with genital epithelial barrier disruption and mucosal influx of human immunodeficiency virus (HIV) target cells. We explored the underlying molecular mechanisms of these findings. Ectocervical biopsies and cervicovaginal lavage (CVL) specimens were collected from HIV-seronegative Kenyan sex workers using DMPA (n=32) or regularly cycling controls (n=64). more...

Oleanolic acid inhibits the development of breast cancer by regulating lactic acid bacteria and the Cytokinecytokine receptor interaction pathway

(Submitter supplied) It was found that after OA treatment, compared with the CK group, the abundance of lactic acid bacteria in the intestinal flora of mice in the OA group increased, and the increase in the abundance of lactic acid bacteria made the gene Il10 upregulated, Il10 had a significant effect on tumor volume reduction and prolongation of mouse survival, and played a role through cytokine receptor interaction pathway.

Transcriptome profiling of the microdissected breast tissue compartments

(Submitter supplied) In addition to the accumulation of pro-oncogenic mutations in the epithelial cells, the tumorigenic process involves the dysregulation of the interactions between the epithelial cells and their microenvironment, as well as alterations within the microenvironment itself. Therefore, investigating the causes of the loss of the normal tissue homeostasis in the earliest stages of breast cancer carcinogenesis is the necessary underpinning of an effective breast cancer prevention strategy. more...
