Changes in PBMC gene expression profiles after influenza vaccination in healthy human subjects

(Submitter supplied) Daily sampling of peripheral blood from human subjects vaccinated for influenza was done immediately before vaccination and for 10 days after vaccination. Temporal patterns of gene expression, determined by RNA-seq, in unfractionated PBMC suggested migration of myeloid/dendritic cell lineage cells one day after vaccination.

Changes in gene expression profiles of circulating B cells after influenza vaccination in healthy human subjects

(Submitter supplied) Daily sampling of peripheral blood from human subjects vaccinated for influenza was done immediately before vaccination and for 10 days after vaccination. In B cells, 90% of transcriptomic variation in subjects who received influenza vaccine within the previous three years was explained by a single temporal pattern unique to the individual. A common set of 742 genes was strongly correlated with the migration of differentiating plasma cell subtypes.

Global Analyses of Human Immune Variation Reveal Baseline Predictors of Postvaccination Responses

(Submitter supplied) A major goal of systems biology is the development of models that accurately predict responses to perturbation. Constructing such models requires the collection of dense measurements of system states, yet transformation of data into predictive constructs remains a challenge. To begin to model human immunity, we analyzed immune parameters in depth both at baseline and in response to influenza vaccination. more...

Time Course of Adults Vaccinated with Influenza TIV Vaccine during 2011/12 Flu Season

(Submitter supplied) Systems approaches have been used to describe molecular signatures driving immunity to influenza vaccination in humans. Whether such signatures are similar across multiple seasons, and in diverse populations is unknown. We applied systems approaches to study immune responses in young and, elderly subjects vaccinated with the seasonal influenza vaccine across 5 consecutive seasons.

A Cell-based Systems Biology Assessment of Human Blood to Monitor Immune Responses After Influenza Vaccination

(Submitter supplied) Systems biology is an approach to comprehensively study complex interactions within a biological system. Most published systems vaccinology studies have utilized whole blood or peripheral blood mononuclear cells (PBMC) to monitor the immune response after vaccination. Because human blood is comprised of multiple hematopoietic cell types, the potential for masking responses of under-represented cell populations is increased when analyzing whole blood or PBMC. more...

Gene expression profiles associated with humoral response to influenza vaccine, Vaxigrip, in young and elderly subjects of Chinese ethnicity in Singapore

(Submitter supplied) While studying influenza vaccine-induced responses, the elderly are typically considered as a homogeneous population. However, chronological age alone is insufficient in defining immune responsiveness of individuals. In this study, a stratified cohort of elderly (65 year old and above) and young (below 35 year old) individuals was recruited to study the heterogeneity of influenza vaccine-induced responses and identify innate signatures including gene expression profiles defining vaccine immunogenicity. more...

Sex-Specific Differences of Humoral Immunity and Transcriptome Diversification in Older Adults Vaccinated with Inactivated Quadrivalent Influenza Vaccines

(Submitter supplied) Clinical data showed sex variability in the immune response to influenza vaccination, this study aimed to investigate differentially expressed genes that contribute to sex-bias immune responses to quadrivalent inactivated influenza vaccines (QIVs) in older subjects. A cohort of 60 healthy older adults aged 60 to 80 years old were vaccinated with quadrivalent inactivated influenza vaccines (QIVs), and gene expression was analyzed at Day 0 pre-vaccination, Day 3, Day 28 and Day 180 post-vaccination. more...
