Selection for contextual fear conditioning affects anxiety-like behaviors and gene expression (palme-affy-mouse-84746)

(Submitter supplied) These data are from the brains (amygdala and hippocampus) of mice originally derived from a cross between C57BL/6J and DBA/2J inbred strains. We used short-term selection to produce outbred mouse lines with differences in contextual fear conditioning, which is a measure of fear learning. We selected for a total of 4 generations. Fear learning differed in the selected lines and this difference was stronger with each successive generation of selection. more...

Classical fear conditioning effect on amygdala and hippocampus

Comparison of amygdalae and hippocampi of animals subjected to classical fear conditioning (CFC), which consists of a 3-minute exposure to the conditioning chamber, a 30-second exposure to tone, and a 2-second, 1.5-mA footshock. CFC induced greater gene expression changes in the amygdala.

Maternal Style Selectively Shapes Amygdalar Development and Social Behavior in Rats Genetically Prone to High Anxiety

(Submitter supplied) The early-life environment critically influences neurodevelopment and later psychological health. To elucidate neural and environmental elements that shape emotional behavior, we developed a rat model of individual differences in temperament and environmental reactivity. We selectively bred rats for high versus low behavioral response to novelty and found that high-reactive (bred high-responder, bHR) rats displayed greater risk-taking, impulsivity and aggression relative to low-reactive (bred low-responder, bLR) rats, which showed high levels of anxiety/depression-like behavior and certain stress vulnerability. more...

Gene Expression Data from Prefrontal Cortex and Nucleus Accumbens from Inbred Strains of Mice

(Submitter supplied) Catechol-O-methyltransferase (COMT) is an ubiquitously expressed enzyme that maintains basic biologic functions by inactivating catechol substrates. In humans, polymorphic variance at the COMT locus has been associated with modulation of pain sensitivity (Andersen & Skorpen, 2009) and risk for developing psychiatric disorders (Harrison & Tunbridge, 2008). A functional haplotype associated with increased pain sensitivity was shown to result in decreased COMT activity by altering mRNA secondary structure-dependent protein translation (Nackley et al., 2006). more...

Gene networks associated with conditional fear in mice identified using a systems genetics approach

(Submitter supplied) To uncover genetic networks governing the biology of conditional fear, we used a systems genetics approach to analyze a hybrid mouse diversity panel (HMDP) with high mapping resolution. By integrating fear phenotypes, transcript profiling data from hippocampus and striatum and genotype information, two gene co-expression networks correlated with context-dependent immobility were identified. We prioritized the key markers and genes in these pathways using intramodular connectivity measures and structural equation modeling. more...

Hippocampal RNA-seq in mice selectively bred for high and low open-field activity (OFA)

(Submitter supplied) Purpose: To investigate the effect that selective breeding for high and low OFA has on gene expression Methods: Male and female High and Low Activity mice (n=4 per strain/sex) were euthanized at postnatal day 70 and hippocampi were immediately collected. Paired-end RNA-seq methods were employed to assess hippocampal mRNA expression. Results: The expression of thousands of transcripts were altered by selective breeding for high and low OFA.
