Transcriptional profiling of THP-1 monocytes conditioned with Lacticaseibacilli rhamnosus R0011 secretome

(Submitter supplied) Macrophage responses to activation are fluid and dynamic in their ability to respond appropriately to challenges, a role integral to host defence. While bacteria can influence macrophage differentiation and polarization into pro-inflammatory and alternatively activated phenotypes through direct interactions, many questions surround indirect communication mechanisms mediated through secretomes derived from gut bacteria, such as lactobacilli. more...

Gene expression changes between lenalidomide (LEN)-sensitive and LEN-resistant human multiple myeloma (MM) cell lines

(Submitter supplied) To investigate the relationship between the LEN resistance and gene expression profile, we generated LEN-resistant (LEN-R) cell clones using U266 and NCU-MM1 cell lines by chronically exposing low dose of LEN (0.1 to 10 μM). The parental and LEN-R MM cells were incubated with either DMSO or LEN (10 μM) for 24 h.

Molecular Markers for Predicting Treatment Outcome in Patients with Rectal Cancer: A Comprehensive Analysis from the German Rectal Cancer Trials

(Submitter supplied) Background: Validated markers to predict outcome in rectal cancer patients treated with multimodal therapy remain elusive. Identifying molecular profiles for disease prognosis would be required for the design of clinical trials aimed at optimizing risk-adapted therapies. We have therefore used whole genome expression profiling of tumors of a large cohort of patients enrolled in the multicenter trials of the German Rectal Cancer Study Group (GRCSG) to identify molecular profiles for individualized therapy. more...

Human Chronic Lymphocytic Leukaemia cells: Untreated vs mithramycin-treated

(Submitter supplied) Transcriptional profiling of primary human malignant B cells from patients with chronic lymphocytic leukaemia (CLL), comparing untreated CLL cells with CLL cells that have been treated 24h with 200nM mithramycin. Mithramycin intercalates into GC-rich regions of DNA to inhibit binding of the transcription factor SP1. The goal of this experiment is to gain understanding of the genes regulated by SP1 in primary CLL cells.

Transcriptomic analysis of HepG2 cells treated with BDE-47, BDE-99, BDE-209 and their ternary mixture at 1nM as a dietary relevant concentration

(Submitter supplied) Polybrominated diphenyl ethers (PBDEs) are persistent organic chemicals implied as flame re-tardants. Humans are mainly exposed to BDE-47, -99 and -209 congeners by diet. PBDEs are metabolic disruptors with liver as main target organ. To investigate their mode of action at a human relevant concentration, we exposed HepG2 cells to these congeners and their mixture at 1 nM for 72h, analyzing their transcriptomic and proteomic profiles. more...

PI3Kβ and β-Catenin mediate EZH2 removal from promoters controlling human ESC stemness and primitive streak gene expression

(Submitter supplied) The mechanism governing the transition of human embryonic stem cells towards differentiated cells is only partially understood. To explore this transition, the activity and expression of the ubiquitous phosphoinositide-3-kinase (PI3K α and β) were modulated. This study reports a pathway that dismantles the repression imposed by the EZH2 polycomb repressor on an essential stemness gene, NODAL, and on transcription factors required at differentiation to trigger primitive streak formation. more...

Transcriptomic heterogeneity of the prostate cancer

(Submitter supplied) The transcriptomic heterogeneity of the prostate cancer was tested by profiling histologically distinct but equally graded (Gleason score 4+5=9/10) cancer nodules from a surgically removed prostate cancer. We found that not only that the genes were differently regulated in the two nodules but also that expression fluctuations were differently controlled and the gene networks differently remodeled.
