Description

1st strand cDNA was primed with a Not I - oligo(dT) primer [5' TGTTACGAATCTGAAGTGGGAGCGGCCGCCCTTTTTTTTTTTTTTTTTTTTTTTTT 3']; double-stranded cDNA was ligated to Eco RI adaptors [AATTCGGATCCTTG], digested with Not I and cloned into the Not I and Eco RI sites of the modified pT7T3 vector. Library constructed by Bob Barstead. The C2C12 cell line (available from ATCC, catalog # CRL-1772) differentiates rapidly, forming contractile myotubes and producing characteristic muscle proteins.
