Total RNA was prepared from the whole Thellungiella plants using Trizol reagent (Gibico, USA). Total RNAs of different time points were mixed and used for further mRNA isolation according to manufacturer's protocol (Qiagen, Germany). Briefly, both first-strand cDNAs were synthesized and digested with Not I. Sal I adaptors were ligated to these digested cDNAs fragments, and then cDNAs of Thellungiella were directionally inserted into the pREP-5N vector.
